Because heterozygosity it the best determination of genetic diversity in a population. If you have more heterozygous organisms, you have more possibilities of different copies of genetic information to flow through said population.

 

Homozygosity cannot be used to measure genetic diversity, because...

 

a) most organisms are homozygous for about 90% of genetic information

b) homozygous organisms will rarely produce a differing offspring

 

So it's defunct in that regard

Heterozygosity measurement in population genetics refers to measuring the fraction of heterozygotes for certain loci in a deme (or population). This is tricky for several reasons. Microsatellites (SSRs) are, for example, known to be very diverse (can consist out of many alleles). Simply looking at heterozygosity would then give you an outcome closer to one, than if you’d look at more conservative regions of the genome. This does not mean that suddenly the population is less diverse. You also miss out on diversity of alleles this way, because one population can have only heterozygous SSR’s but each SSR could be biallelic for these loci, while another population has heterozygous SSR’s and one homozygous SSR. Measuring heterozygosity would lead to a higher number for the first population, while for all we know the second population might have heterozygous SSR’s consisting out of many alleles, what would make this population more diverse - in that respect - than the first.

There’s more reasons why measuring heterozygosity is not the same as measuring diversity, but people just read a Wikipedia entry and think they know. Sadly peer-reviewed articles nowadays published using heterozygosity as a measurement (eg. Fst, Gst, Rst) for diversity are still more common than the ones using more correct measurements such as Jost D. So I was just wondering why geneticists are still making this (essential) mistake when wanting to measure diversity of demes (subpopulations).
